Culture remains irreplaceable: it recovers viable organisms, proves they are alive, enables colony-based identification and — critically — provides the isolate without which phenotypic antimicrobial susceptibility testing is impossible.
Microbiology is the study of microorganisms — bacteria, viruses, fungi and protozoa — their morphology, physiology, pathogenesis and clinical significance. Culture is its oldest and still most versatile tool.
Microscopy (Gram, AFB stains), inoculation onto selective and enriched media, incubation, colony morphology assessment, and biochemical identification form the classical pathway.
